The M25 anti-clockwise is closed between J4 and J3, due to a collision, Highways England has said.
Emergency services, including Kent Police, are in attendance and drivers are being warned of likely delays on the approach to the closure, along with longer journey times on diversion routes.
"Traffic is being diverted to follow the hollow square symbol, to leave at Junction 4 and return at Junction 3," Highways England said on its website.
"Head via the A224 towards Orpington, and the A20 towards Dover, to return to the M25 at the Swanley Interchange," it advised drivers.
The road is expected to clear between 05:00 and 05:15 today, August 18, and normal traffic conditions expected to resume within the same time frame.
